Improving Business Model Configuration through a Question-based Approach
In the competitive context of agile innovation cycles, it is necessary for companies to construct their business model leading them to a creative strategy innovation. There already are a number of methods to create business models, many of which are also implemented in software. However, these are often unstructured, unguided and static, resulting in diverse and heterogeneous business models. This complicates automated evaluation allowing recommendations. The aim is to develop a question-based tool yielding for comparable and, thus, analyzable business models based on a developed standardized taxonomy. The questions guiding through the configurator were derived from this taxonomy. A tool was developed implementing the question-based concept. User tests were conducted as part of an evaluation showing promising results concerning usability in addition to the already achieved standardization
From OCEL to DOCEL -- Datasets and Automated Transformation
Object-centric event data represent processes from the point of view of all
the involved object types. This perspective has gained interest in recent years
as it supports the analysis of processes that previously could not be
adequately captured, due to the lack of a clear case notion as well as an
increasing amount of output data that needs to be stored. Although publicly
available event logs are crucial artifacts for researchers to develop and
evaluate novel process mining techniques, the currently available
object-centric event logs have limitations in this regard. Specifically, they
mainly focus on control-flow and rarely contain objects with attributes that
change over time, even though this is not realistic, as the attribute values of
objects can be altered during their lifecycle. This paper addresses this gap by
providing two means of establishing object-centric datasets with dynamically
evolving attributes. First, we provide event log generators, which allow
researchers to generate customized, artificial logs with dynamic attributes in
the recently proposed DOCEL format. Second, we propose and evaluate an
algorithm to convert OCEL logs into DOCEL logs, which involves the detection of
event attributes that capture evolving object information and the creation of
dynamic attributes from these. Through these contributions, this paper supports
the advancement of object-centric process analysis by providing researchers
with new means to obtain relevant data to use during the development of new
techniques
Characterization of the first potent and selective PDE9 inhibitor using a cGMP reporter cell
ABSTRACT We report here the in vitro characterization of 1-(2-chlorophenyl)-6-[(2R)-3,3,3-trifluoro-2-methylpropyl]-1,5-dihydro-4H-pyrazolo [3,4-d]pyrimidine-4-one , the first potent and selective inhibitor of phosphodiesterase 9 (PDE9), which is currently under preclinical development for the treatment of Alzheimer's disease. This compound selectively inhibits human (IC 50 Ï 55 nM) and murine (IC 50 Ï 100 nM) PDE9 activity in vitro and shows only moderate activity against other cyclic nucleotide-specific phosphodiesterases. We also report the generation and characterization of a stably transfected PDE9 Chinese hamster ovary cell line, additionally expressing soluble guanylate cyclase (sGC), the olfactory cyclic nucleotide-gated cation channel CNGA2 and the photoprotein aequorin. In this cell line, intracellular cGMP levels can be monitored in real-time via aequorin luminescence induced by Ca 2Ï© influx through CNGA2, acting as the intracellular cGMP sensor. This simple and sensitive assay system was used for the characterization of the cellular activity of the new PDE9 inhibitor. BAY 73-6691 alone did not significantly increase basal cGMP levels in this experimental setting. However, in combination with submaximal stimulating concentrations of the sGC activator 4-[((4-carboxybutyl){2-[(4-phenethylbenzyl)oxy]phenethyl}amino)methyl] benzoic acid (BAY 58-2667), the compound induced concentration-dependent luminescence signals and intracellular cGMP accumulation. The PDE9 inhibitor significantly potentiated the cGMP signals generated by sGC activating compounds such as BAY 58-2667 or 5-cyclopropyl-2-[1-(2-fluorobenzyl)-1H-pyrazolo [3,4-b]pyridin-3-yl]pyrimidin-4-ylamine (BAY 41-2272) and induced leftward shifts of the corresponding concentration-response curves. Using our newly generated PDE9 reporter cell line, we could show that BAY 73-6691 is able to efficiently penetrate cells and to inhibit intracellular PDE9 activity
Large Process Models: Business Process Management in the Age of Generative AI
The continued success of Large Language Models (LLMs) and other generative
artificial intelligence approaches highlights the advantages that large
information corpora can have over rigidly defined symbolic models, but also
serves as a proof-point of the challenges that purely statistics-based
approaches have in terms of safety and trustworthiness. As a framework for
contextualizing the potential, as well as the limitations of LLMs and other
foundation model-based technologies, we propose the concept of a Large Process
Model (LPM) that combines the correlation power of LLMs with the analytical
precision and reliability of knowledge-based systems and automated reasoning
approaches. LPMs are envisioned to directly utilize the wealth of process
management experience that experts have accumulated, as well as process
performance data of organizations with diverse characteristics, e.g., regarding
size, region, or industry. In this vision, the proposed LPM would allow
organizations to receive context-specific (tailored) process and other business
models, analytical deep-dives, and improvement recommendations. As such, they
would allow to substantially decrease the time and effort required for business
transformation, while also allowing for deeper, more impactful, and more
actionable insights than previously possible. We argue that implementing an LPM
is feasible, but also highlight limitations and research challenges that need
to be solved to implement particular aspects of the LPM vision
Augmented serum level of major histocompatibility complex class I-related chain A (MICA) protein and reduced NKG2D expression on NK and T cells in patients with cervical cancer and precursor lesions
<p>Abstract</p> <p>Background</p> <p>Cervical cancer is the second most common cancer in women worldwide. NK and cytotoxic T cells play an important role in the elimination of virus-infected and tumor cells through NKG2D activating receptors, which can promote the lysis of target cells by binding to the major histocompatibility complex class I-related chain A (MICA) proteins. Increased serum levels of MICA have been found in patients with epithelial tumors. The aim of this study was to compare the levels of soluble MICA (sMICA) and NKG2D-expressing NK and T cells in blood samples from patients with cervical cancer or precursor lesions with those from healthy donors.</p> <p>Methods</p> <p>Peripheral blood with or without heparin was collected to obtain mononuclear cells or sera, respectively. Serum sMICA levels were measured by ELISA and NKG2D-expressing immune cells were analyzed by flow cytometry. Also, a correlation analysis was performed to associate sMICA levels with either NKG2D expression or with the stage of the lesion.</p> <p>Results</p> <p>Significant amounts of sMICA were detected in sera from nearly all patients. We found a decrease in the number of NKG2D-expressing NK and T cells in both cervical cancer and lesion groups when compared to healthy donors. Pearson analysis showed a negative correlation between sMICA and NKG2D-expressing T cells; however, we did not find a significant correlation when the analysis was applied to sMICA and NKG2D expression on NK cells.</p> <p>Conclusion</p> <p>Our results show for the first time that high sMICA levels are found in sera from patients with both cervical cancer and precursor lesions when compared with healthy donors. We also observed a diminution in the number of NKG2D-expressing NK and T cells in the patient samples; however, a significant negative correlation between sMICA and NKG2D expression was only seen in T cells.</p
